A Morphological Study on the Cytotoxicity of Several Anticancer Drugs (Adriamycin, Cytosine arabinoside and Vinblastine) on Cultured Mouse Fibroblasts



Abstract
To investigate to cytotoxic effects of adriamycin, cytosine arabinoside and vinblastine on cultured mouse fibroblasts, colorimetric assays of neutral red(NR) and tetrazolium MTT, protein content and electron microscopic studies were performed.
Adriamycin, cytosine arabinoside and vinblastine were highly toxic by showing that values of NR50 and MTT50 of each drug were lower than a concentration of 100¥ìM, respectively.
The amount of total protein in anticancer drug-treated groups were less than those of the control. The control. The number of cells grown in anticancer drug-treated groups were less than those of the control.
Electron microscopic study showed that fibroblasts treated with adriamycin showed increment of lysosome and vacuoles as well as cisternal dilatation of rough endoplasmic reticulum, and cytosine arabinoside treated cells had nucleus with
fragmental
heterochromatin, swollen mitochondria and many vacuoles, and vinblastine treated cells contained elongated and often lobulated nucleus and many cytoplasmic vacuoles.
These reults suggest that adriamycin, cytosine arabinoside and vinblastine have severe cytotoxic effects on the cultured mouse fibroblasts by inhibition of protein synthesis and structural changes of cell organelles.
